The BioAssay ELISA Kit is a 1.5 hour solid-phase competitive ELISA designed for the quantitative determination of Bone Alkaline Phosphatase (BALP) in rat serum, plasma, cell culture supernatant, tissue homogenates and body fluids. This kit is for research use only, not for therapeutic or diagnostic applications.Assay Principle:Bone Alkaline Phosphatase (BALP) BioAssay ELISA Kit (Rat) utilizes the quantitative Competitive Enzyme Immunoassay technique, utilizing a monoclonal antibody and a conjugate to Bone Alkaline Phosphatase (BALP) (Rat). The assay sample and buffer are incubated together with the HRP conjugate in a pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Target Protein concentration since Protein from samples and Protein-HRP conjugate compete for the anti-Protein antibody binding site. Since the number of sites is limited, as more sites are occupied by Protein from the sample, fewer sites are left to bind the Protein-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Target Protein concentration in each sample is interpolated from this standard curve.Detection Range:0.5-10ng/mlSensitivity:0.1ng/mlPrecision:Intra-Assay %CV: <10Inter-Assay %CV: <10Specificity:This assay has high sensitivity and excellent specificity for detection of Bone Alkaline Phosphatase (BALP). No significant crossreactivity or interference with analogs was observed. Note: Crossreactivity detection between BALP and all analogs has not been determined and crossreaction may exist in some cases.Kit Components:183576A: Microtiter Strips, 1x48 wells or 1x96 wells.
