Circulating Immune Complexes (CIC) BioAssay ELISA Kit is a 1.5-hour solid-phase competitive ELISA designed for the quantitative determination of CIC in rat serum, plasma, tissue homogenates and body fluids. This kit is intended for research use only and may not be used for therapeutic or diagnostic applications.Detection Range:1-25ug/mlSensitivity:0.1ug/mlPrecision:Intra-assay %CV: <10Inter-assay %CV: <10Specificity: This assay has high sensitivity and excellent specificity for detection of CIC (Rat). No significant crossreactivity or interference with analogs was observed. Note: Crossreactivity detection between the CIC and all analogs has not been determined and crossreaction may exist in some cases.Kit Components:186806A: Microtiter Strips, 1x48 wells or 1x96 wells186806B: Enzyme Conjugate, 1x3ml or 1x6ml186806C: Standard, 0ug/ml, 1x1 vial186806D: Standard, 1ug/ml, 1x1 vial186806E: Standard, 2.5ug/ml, 1x1 vial186806F: Standard, 5ug/ml, 1x1 vial186806G: Standard, 10ug/ml, 1x1 vial186806H: Standard, 25ug/ml, 1x1 vial186806J: Substrate A, 1x6ml186806K: Substrate B, 1x6ml186806L: Stop Solution, 1x6ml186806M: Wash Solution (100X), 1x10ml186806N: Balance Solution, 1x3mlNote:The BALANCE SOLUTION is used only when the sample is cell culture supernatants, body fluid and tissue homogenate; if the sample is serum or plasma, then the BALANCE SOLUTION is a superfluous reagent. Storage and Stability:Store components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap.Assay Principle:Circulating Immune Complexes (CIC) BioAssay ELISA Kit employs the quantitative competitive enzyme-linked immunoassay technique, utilizing a microtiter plate that is has been pre-coated with a monoclonal anti-CIC antibody. The assay samples and standards are incubated together with CIC-HRP conjugate for one hour. A competition for limited antibody binding sites on the plate occurs between CIC-HRP conjugate and CIC in the samples and standards. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to terminate the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CIC concentration in the sample or standard. A standard curve is plotted relating the intensity of the color (O.D.) to the concentrations of the standards. The CIC concentration in each sample is interpolated from this standard curve.Contact:United States Biological4 Technology WaySalem, MA 01970www.usbio.netTech Support: tech@usbio.netCustomer Service: service@usbio.netTel: 781-639-5092 • 800-520-3011Fax: 978-594-8052
