Glutathione Peroxidase (GSH-Px) BioAssay ELISA Kit (Rat) is a 1.5-hour solid-phase competitive ELISA designed for the quantitative determination of GSH-Px in Rat serum, plasma, tissue homogenates and body fluids. This kit is intended for research use only and may not be used for therapeutic or diagnostic applications.Detection Range: 5-100ng/mlSensitivity: 1ng/mlPrecision:Intra-assay %CV: <10Inter-assay %CV: <10Specificity: This assay has high sensitivity and excellent specificity for detection of GSH-Px (Rat). No significant crossreactivity or interference with analogs was observed.Note: Crossreactivity detection between the GSH-Px and all analogs has not been determined and crossreaction may exist in some cases.Kit Components:193180A: Microtiter Strips, 1x48 wells or 1x96 wells193180B: Enzyme Conjugate, 1x3ml or 1x6ml193180C: Standard, 0ng/ml, 1x1 vial193180D: Standard, 5ng/ml, 1x1 vial193180E: Standard, 10ng/ml, 1x1 vial193180F: Standard, 25ng/ml, 1x1 vial193180G: Standard, 50ng/ml, 1x1 vial193180H: Standard, 100ng/ml, 1x1 vial193180J: Substrate A, 1x6ml193180K: Substrate B, 1x6ml193180L: Stop Solution, 1x6ml193180M: Wash Solution (100X), 1x10ml193180N: Balance Solution, 1x3mlStorage and Stability:Store components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap.Assay Principle:The Glutathione Peroxidase (GSH-Px) BioAssay ELISA Kit (Rat) employs the quantitative competitive enzyme-linked immunoassay technique, utilizing a microtiter plate that is has been pre-coated with a monoclonal anti-GSH-Px antibody. The assay samples and standards are incubated together with GSH-Px-HRP conjugate for one hour. A competition for limited antibody binding sites on the plate occurs between GSH-Px-HRP conjugate and GSH-Px in the samples and standards. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to terminate the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GSH-Px concentration in the sample or standard. A standard curve is plotted relating the intensity of the color (O.D.) to the concentrations of the standards. The GSH-Px concentration in each sample is interpolated from this standard curve.Contact:United States Biological4 Technology WaySalem, MA 01970www.usbio.netTech Support: tech@usbio.netCustomer Service: service@usbio.netTel: 781-639-5092 • 800-520-3011Fax: 978-594-8052
