The BioAssay ELISA Kit is a 1.5 hour solid-phase competitive ELISA designed for the quantitative determination of Heat Shock Protein 70 (HSP70) in Rat serum, plasma, cell culture supernatants, tissue homogenates and other body fluids. This kit for research use only, not for therapeutic or diagnostic applications.Detection Range: 0.5-10ng/mlSensitivity: 0.1ng/mlPrecision:Intra-assay CV%: 5.9, 8.2Inter-assay CV%: 4.5, 6.2Specificity: This assay has high sensitivity and excellent specificity for detection of HSP70 (Rat). No significant crossreactivity or interference with analogs was observed.Note: Crossreactivity detection between the HSP70 and all analogs has not been determined and crossreaction may exist in some cases.Kit Components:193797A: Microtiter Strips, 1x48 wells or 1x96 wells193797B: Enzyme Conjugate, 1x3ml or 1x6ml193797C: Standard, 0ng/ml, 1x1 vial193797D: Standard, 0.5ng/ml, 1x1 vial193797E: Standard, 1ng/ml, 1x1 vial193797F: Standard, 2.5ng/ml, 1x1 vial193797G: Standard, 5ng/ml, 1x1 vial193797H: Standard, 10ng/ml, 1x1 vial193797J: Substrate A, 1x6ml193797K: Substrate B, 1x6ml193797L: Stop Solution, 1x6ml193797M: Wash Solution (100X), 1x10ml193797N: Balance Solution, 1x3mlNote:The BALANCE SOLUTION is used only when the sample is cell culture supernatants, body fluid and tissue homogenate; if the sample is serum or plasma, then the BALANCE SOLUTION is a superfluous reagent. Storage and Stability:Store components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap.Assay Principle:The Heat Shock Protein 70 (HSP70) BioAssay ELISA kit employs the quantitative competitive enzyme-linked immunoassay technique, utilizing a microtiter plate that is has been pre-coated with a monoclonal anti-HSP70 antibody. The assay samples and standards are incubated together with HSP70-HRP conjugate for one hour. A competition for limited antibody binding sites on the plate occurs between HSP70-HRP conjugate and HSP70 in the samples and standards. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to terminate the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HSP70 concentration in the sample or standard. A standard curve is plotted relating the intensity of the color (O.D.) to the concentrations of the standards. The HSP70 concentration in each sample is interpolated from this standard curve.Contact:United States Biological4 Technology WaySalem, MA 01970www.usbio.netTech Support: tech@usbio.netCustomer Service: service@usbio.netTel: 781-639-5092 • 800-520-3011Fax: 978-594-8052
