Mast Cell Tryptase (MCT) BioAssay ELISA Kit (Mouse) is a 1.5 hour solid-phase Sandwich ELISA for the quantitative determination of Mast Cell Tryptase in mouse serum, plasma, cell culture supernatants, body fluid and tissue homogenates.Detection Range:100-2500pg/mlSensitivity:1pg/mlPrecision:Intra-Assay CV: <10%Inter-Assay CV: <10%Specificity: This assay has high sensitivity and excellent specificity for detection of MCT (Mouse). No significant crossreactivity or interference with analogs was observed. Note: Crossreactivity detection between the MCT (Mouse) and all analogs has not been determined and crossreaction may exist in some cases.Kit Components:196453A: Microtiter Strips, 1x48 or 1x96 wells196453B: Enzyme Conjugate, 1x10ml196453C: Standard, 0pg/ml, 1x1 vial196453D: Standard, 100pg/ml, 1x1 vial196453E: Standard, 250pg/ml, 1x1 vial196453F: Standard, 500pg/ml, 1x1 vial196453G: Standard, 1000pg/ml, 1x1 vial196453H: Standard, 2500pg/ml, 1x1 vial196453J: Substrate A, 1x6ml196453K: Substrate B, 1x6ml196453L: Stop Solution, 1x6ml196453M: Wash Solution (100X), 1x10ml196453N: Balance Solution, 1x3mlStorage and Stability:Store components at 4°C. The microtiter plate has been pre-coated with a monoclonal antibody specific for Mast Cell Tryptase (MCT) (Mouse). Standards or samples are then added to the microtiter plate wells and if the protein is present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of protein present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for the protein are added to each well to “sandwich” the protein immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain the target protein and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulfuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The protein concentration in each sample is interpolated from this standard curve.Contact:United States Biological4 Technology WaySalem, MA 01970www.usbio.net
