The BioAssay ELISA Kit is a 1.5 hour solid-phase sandwich ELISA designed for the quantitative determination of Myelin Basic Protein (MBP) in rat serum, plasma, cell culture supernatants, tissue homogenates and other body fluids. This kit for research use only, not for therapeutic or diagnostic applications.Range: 0.5-10ng/mlSensitivity: 0.1ng/mlPrecision:Intra-assay CV%: 6.3, 8.1Inter-assay CV%: 5.7, 7.2Specificity: This assay has high sensitivity and excellent specificity for detection of MBP (Rat). No significant crossreactivity or interference with analogs was observed.Note: Crossreactivity detection between the MBP (Rat) and all analogs has not been determined and crossreaction may exist in some cases.Kit Components:197274A: Microtiter Strips, 1x48 or 1x96 wells197274B: Enzyme Conjugate, 1x3ml or 1x10ml197274C: Standard, 0ng/ml, 1x1 vial197274D: Standard, 0.5ng/ml, 1x1 vial197274E: Standard, 1ng/ml, 1x1 vial197274F: Standard, 2.5ng/ml, 1x1 vial197274G: Standard, 5ng/ml, 1x1 vial197274H: Standard, 10ng/ml, 1x1 vial197274J: Substrate A, 1x6ml197274K: Substrate B, 1x6ml197274L: Stop Solution, 1x6ml197274M: Wash Solution (100X), 1x10ml197274N: Balance Solution, 1x3mlNote:The BALANCE SOLUTION is used only when the sample is cell culture supernatants, body fluid and tissue homogenate; if the sample is serum or plasma, then the BALANCE SOLUTION is a superfluous reagent. Storage and Stability:Store components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap.Assay Principle:Myelin Basic Protein (MBP) BioAssay ELISA Kit (Rat) utilizes the quantitative Sandwich Enzyme Immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for MBP (Rat). Standards or samples are then added to the microtiter plate wells and if the protein is present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of protein present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for the protein are added to each well to “sandwich” the protein immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain the target protein and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulfuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The protein concentration in each sample is interpolated from this standard curve.Contact:United States Biological4 Technology WaySalem, MA 01970www.usbio.netTech Support: tech@usbio.netCustomer Service: service@usbio.netTel: 781-639-5092 • 800-520-3011Fax: 978-594-8052
