产品描述: Assay Principle:Osteocalcin/Bone gla Protein (OT/BGP) BioAssay ELISA Kit (Rat) utilizes the quantitative Competitive Enzyme Immunoassay technique, utilizing a monoclonal antibody and a conjugate to Osteocalcin/Bone gla Protein (OT/BGP) (Rat). The assay sample and buffer are incubated together with the HRP conjugate in a pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Target Protein concentration since Protein from samples and Protein-HRP conjugate compete for the anti-Protein antibody binding site. Since the number of sites is limited, as more sites are occupied by Protein from the sample, fewer sites are left to bind the Protein-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Target Protein concentration in each sample is interpolated from this standard curve.Intended Use:The BioAssay ELISA Kit is a 1.5 hour solid-phase competitive ELISA designed for the quantitative determination of Osteocalcin/Bone gla Protein (OT/BGP) in rat serum, plasma, cell culture supernatant, tissue homogenates and body fluids. This kit is for research use only, not for therapeutic or diagnostic applications.Detection Range:0.5-10ng/mlSensitivity:0.1ng/mlPrecision:Intra-Assay %CV: 7.5, 8.8Inter-Assay %CV: 5.7, 6.7Specificity:This assay has high sensitivity and excellent specificity for detection of Osteocalcin/Bone gla Protein (OT/BGP).
