The BioAssay ELISA Kit is a 1.5 hour solid-phase Sandwich ELISA designed for the quantitative determination of Osteoprotegerin (OPG) in rat serum, plasma, cell culture supernatants, tissue homogenates and other body fluids. This kit for research use only, not for therapeutic or diagnostic applicationsDetection Range:0.5-10ng/mlSensitivity:0.1ng/mlPrecision:Intra-Assay %CV: 5.2, 7.8Inter-Assay %CV: 4.2, 5.5Specificity:This assay has high sensitivity and excellent specificity for detection of Osteoprotegerin (OPG) (Rat). No significant crossreactivity or interference with analogs was observed. Note: Crossreactivity detection between the Target Protein and all analogs has not been determined and crossreaction may exist in some cases.Assay Principle:Osteoprotegerin (OPG) BioAssay ELISA Kit (Rat) utilizes the quantitative Sandwich Enzyme Immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for rat Osteoprotegerin. Standards or samples are then added to the microtiter plate wells and if the protein is present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of OPG present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for the OPG are added to each well to “sandwich” the protein immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain the OPG and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulfuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The protein concentration in each sample is interpolated from this standard curve.Kit Components:200817A: Microtiter Strips, 1x96 wells 200817B: Enzyme Conjugate, 1x10ml200817C: Standard, 0ng/ml, 1x1 vial200817D: Standard, 0.5ng/ml, 1x1 vial200817E: Standard, 1.0ng/ml, 1x1 vial200817F: Standard, 2.5ng/ml, 1x1 vial200817G: Standard, 5.0ng/ml, 1x1 vial200817H: Standard, 10ng/ml, 1x1 vial200817J: Substrate A, 1x6ml200817K: Substrate B, 1x6ml200817L: Stop Solution, 1x6ml200817M: Wash Solution, 100X, 1x10ml200817N: Balance Solution, 1x3mlStorage and Stability:Store components at 4°C.
