The BioAssay ELISA Kit is a 1.5 hour solid-phase sandwich ELISA designed for the quantitative determination of p53 in rat serum, plasma, cell lysates, cell culture supernatants, tissue homogenates and other body fluids. This kit for research use only, not for therapeutic or diagnostic applications.Detection Range: 2.5-50ng/mlSensitivity: 0.1ng/mlPrecision:Intra-assay CV%: 5.2, 5.8Inter-assay CV%: 3.7, 4.9Specificity: This assay has high sensitivity and excellent specificity for detection of p53 (Rat). No significant crossreactivity or interference with analogs was observed.Note: Crossreactivity detection between the p53 (Rat) and all analogs has not been determined and crossreaction may exist in some cases.Kit Components:200907A: Microtiter Strips, 1x48 or 1x96 wells200907B: Enzyme Conjugate, 1x3ml or 1x10ml200907C: Standard, 0ng/ml, 1x1 vial200907D: Standard, 2.5ng/ml, 1x1 vial200907E: Standard, 5ng/ml, 1x1 vial200907F: Standard, 10ng/ml, 1x1 vial200907G: Standard, 25ng/ml, 1x1 vial200907H: Standard, 50ng/ml, 1x1 vial200907J: Substrate A, 1x6ml200907K: Substrate B, 1x6ml200907L: Stop Solution, 1x6ml200907M: Wash Solution (100X), 1x10ml200907N: Balance Solution, 1x3mlNote:The BALANCE SOLUTION is used only when the sample is cell culture supernatants, body fluid and tissue homogenate; if the sample is serum or plasma, then the BALANCE SOLUTION is a superfluous reagent. Storage and Stability:Store components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap.Assay Principle:The p53 BioAssay ELISA Kit (Rat) utilizes the quantitative Sandwich Enzyme Immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for p53 (Rat). Standards or samples are then added to the microtiter plate wells and if the protein is present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of protein present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for the protein are added to each well to “sandwich” the protein immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain the target protein and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulfuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The protein concentration in each sample is interpolated from this standard curve.Contact:United States Biological4 Technology WaySalem, MA 01970www.usbio.netTech Support: tech@usbio.netCustomer Service: service@usbio.netTel: 781-639-5092 • 800-520-3011Fax: 978-594-8052
