The BioAssay ELISA Kit is a 1.5 hour solid-phase competitive ELISA designed for the quantitative determination of Peptidoglycan in Rat serum, plasma, cell culture supernatants, tissue homogenates and other body fluids. This kit for research use only, not for therapeutic or diagnostic applications.Detection Range:1-25ng/mlSensitivity: 0.1ng/mlPrecision:Intra-assay CV%: 5.9, 7.6Inter-assay CV%: 5.3, 6.7Specificity:This assay has high sensitivity and excellent specificity for detection of Peptidoglycan (Rat). No significant crossreactivity or interference with analogs was observed.Note: Crossreactivity detection between the Peptidoglycan and all analogs has not been determined and crossreaction may exist in some cases.Kit Components:201096A: Microtiter Strips, 1x48 wells or 1x96 wells201096B: Enzyme Conjugate, 1x3ml or 1x6ml201096C: Standard, 0ng/ml, 1x1 vial201096D: Standard, 1ng/ml, 1x1 vial201096E: Standard, 2.5ng/ml, 1x1 vial201096F: Standard, 5ng/ml, 1x1 vial201096G: Standard, 10ng/ml, 1x1 vial201096H: Standard, 25ng/ml, 1x1 vial201096J: Substrate A, 1x6ml201096K: Substrate B, 1x6ml201096L: Stop Solution, 1x6ml201096M: Wash Solution (100X), 1x10ml201096N: Balance Solution, 1x3mlNote:The BALANCE SOLUTION is used only when the sample is cell culture supernatants, body fluid and tissue homogenate; if the sample is serum or plasma, then the BALANCE SOLUTION is a superfluous reagent. Storage and Stability:Store components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap.Assay Principle:This Peptidoglycan BioAssay ELISA Kit employs the quantitative competitive enzyme-linked immunoassay technique, utilizing a microtiter plate that is has been pre-coated with a monoclonal anti-Peptidoglycan antibody. The assay samples and standards are incubated together with Peptidoglycan-HRP conjugate for one hour. A competition for limited antibody binding sites on the plate occurs between Peptidoglycan-HRP conjugate and Peptidoglycan in the samples and standards. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to terminate the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Peptidoglycan concentration in the sample or standard. A standard curve is plotted relating the intensity of the color (O.D.) to the concentrations of the standards. The Peptidoglycan concentration in each sample is interpolated from this standard curve.Contact:United States Biological4 Technology WaySalem, MA 01970www.usbio.netTech Support: tech@usbio.netCustomer Service: service@usbio.netTel: 781-639-5092 • 800-520-3011Fax: 978-594-8052
