Plasma Kallikrein (KLKB1) BioAssay ELISA Kit is a 1.5 hour solid-phase competitive ELISA designed for the quantitative determination of Plasma KLKB1 in rat serum, plasma, cell culture supernatant, tissue homogenates and body fluids. This kit is intended for research use only and may not be used for therapeutic or diagnostic applications.Detection Range:50-1000pg/mlSensitivity:1pg/mlPrecision:Intra-Assay %CV: <10Inter-Assay %CV: <10Specificity:This assay has high sensitivity and excellent specificity for detection of KLKB1 (Rat). No significant crossreactivity or interference with analogs was observed. Note: Crossreactivity detection between KLKB1 and all analogs has not been determined and crossreaction may exist in some cases.Kit Components:201486A: Microtiter Strips, 1x48 wells or 1x96 wells201486B: Enzyme Conjugate, 1x3ml or 1x6ml201486C: Standard, 0pg/ml, 1x1 vial201486D: Standard, 50pg/ml, 1x1 vial201486E: Standard, 100pg/ml, 1x1 vial201486F: Standard, 250pg/ml, 1x1 vial201486G: Standard, 500pg/ml, 1x1 vial201486H: Standard, 1000pg/ml, 1x1 vial201486J: Substrate A, 1x6ml201486K: Substrate B, 1x6ml201486L: Stop Solution, 1x6ml201486M: Wash Solution, 100X, 1x10ml201486N: Balance Solution, 1x3mlNote:The BALANCE SOLUTION is used only when the sample is cell culture supernatants, body fluid and tissue homogenate; if the sample is serum or plasma, then the BALANCE SOLUTION is a superfluous reagent. Storage and Stability:Store components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Assay Principle:This ELISA kit utilizes the quantitative Competitive Enzyme Immunoassay technique, utilizing a monoclonal antibody anti-KLKB1 and a KLKB1-HRP conjugate. The assay sample and buffer are incubated together with the HRP conjugate in a pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the KLKB1 concentration since KLKB1 from samples and KLKB1-HRP conjugate compete for the anti-KLKB1 antibody binding site. Since the number of sites is limited, as more sites are occupied by KLKB1 from the sample, fewer sites are left to bind the KLKB1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The KLKB1 concentration in each sample is interpolated from this standard curve.Contact:United States Biological4 Technology WaySalem, MA 01970www.usbio.netTech Support: tech@usbio.netCustomer Service: service@usbio.netTel: 781-639-5092 • 800-520-3011Fax: 978-594-8052
