Superoxide Dismutase (SOD) BioAssay ELISA Kit (Rat) is a 1.5-hour solid-phase competitive ELISA designed for the quantitative determination of SOD in rat serum, plasma, cell culture supernatants, tissue homogenates and body fluids. This kit is intended for research use only and may not be used for therapeutic or diagnostic applications.Detection Range: 1-25ug/mlSensitivity: 0.1ug/mlPrecision:Intra-assay %CV: <10Inter-assay %CV: <10Specificity: This assay has high sensitivity and excellent specificity for detection of SOD (Rat). No significant crossreactivity or interference with analogs was observed. Note: Crossreactivity detection between the SOD and all analogs has not been determined and crossreaction may exist in some cases.Kit Components:204427A: Microtiter Strips, 1x48 wells or 1x96 wells204427B: Enzyme Conjugate, 1x3ml or 1x6ml204427C: Standard, 0ug/ml, 1x1 vial204427D: Standard, 1ug/ml, 1x1 vial204427E: Standard, 2.5ug/ml, 1x1 vial204427F: Standard, 5ug/ml, 1x1 vial204427G: Standard, 10ug/ml, 1x1 vial204427H: Standard, 25ug/ml, 1x1 vial204427J: Substrate A, 1x6ml204427K: Substrate B, 1x6ml204427L: Stop Solution, 1x6ml204427M: Wash Solution (100X), 1x10ml204427N: Balance Solution, 1x3mlStorage and Stability:Store components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap.Assay Principle:Superoxide Dismutase (SOD) BioAssay ELISA Kit (Rat) employs the quantitative competitive enzyme-linked immunoassay technique, utilizing a microtiter plate that is has been pre-coated with a monoclonal anti-SOD antibody. The assay samples and standards are incubated together with SOD-HRP conjugate for one hour. A competition for limited antibody binding sites on the plate occurs between SOD-HRP conjugate and SOD in the samples and standards. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to terminate the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SOD concentration in the sample or standard. A standard curve is plotted relating the intensity of the color (O.D.) to the concentrations of the standards. The SOD concentration in each sample is interpolated from this standard curve.Contact:United States Biological4 Technology WaySalem, MA 01970www.usbio.netTech Support: tech@usbio.netCustomer Service: service@usbio.netTel: 781-639-5092 • 800-520-3011Fax: 978-594-8052
