The rat IL-1 beta BioAssay ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of rat IL-1 beta.Sample Type:Serum, plasma and cell culture supernatantsIntended Use:For use in quantitative protein detection, establishing normal range and validation of antibody array results.Sensitivity:80pg/mlRange:80-50,000pg/mlSpecificity:Recognizes rat IL-1 beta. This ELISA kit shows no cross-reactivity with the following cytokines tested: rat CINC-2, CINC-3, CNTF, Fractalkine, IL-1a, IL-1b, IL-4, IL-6, IL-10, GM-CSF, IFN-g, Leptin, Lix, MCP-1, MIP-3a, b-NGF, TIMP-1, TNF-a.Test Principle:This BioAssay employs an antibody specific for IL-1 beta coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-1 beta present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-rat IL-1 beta antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-1 beta bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450nm. The intra-Assay reproducibility is CV<10% & inter-Assay is CV<12%.Kit Components:IL-1 beta Coated Microplate (Item A), 1x96wells 20X Wash Buffer Concentrate (Item B), 1x25mlStandards (Item C) (recombinant IL-1 beta), 2x vialsAssay Diluent A (Item D) for Standard/Sample (serum/plasma), 1x30ml Assay Diluent B (Item E) for Standard/Sample (cell culture medium), 5x conc., 1x15ml Detection Antibody IL-1 beta (Item F), 2x vial HRP-Streptavidin Concentrate (Item G), 200x concentrated, 1x200ul TMB One-Step Substrate Reagent (Item H), 1x12ml Stop Solution (Item I), 0.2M sulfuric acid, 1x8mlStorage and Stability:Store powder at 4°C liquid at -20°C.
