The rat MMP-8 BioAssay ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of rat MMP-8. Sample Type:Serum and plasma (using heparin as an anticoagulant, EDTA and citrate are not recommended). Cell culture supernatants and urine.Intended Use:Applications include: quantitative protein detection, establishing normal range and validation of antibody array results. Sensitivity:The minimum detectable dose of MMP-8 is typically <50pg/ml. Specificity:Recognizes rat MMP-8. This ELISA kit shows no cross-reactivity with the following cytokines tested: rat CINC-2, CINC-3, CNTF, Fractalkine, IL-1alpha, IL-1beta, IL-4, IL-6, IL-10, GM-CSF, IFN-gamma, Leptin, Lix, MCP-1, MIP-3alpha, beta-NGF, TIMP-1, TNF-alpha.Test Principle:This BioAssay employs an antibody specific for MMP-8 coated on a 96-well plate. Standards and samples are pipetted into the wells and MMP-8 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Rat MMP-8 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of MMP-8 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450nm. The intra-Assay reproducibility is CV<10% and inter-Assay is CV<12%.Kit Components:MMP-8 Microplate (Item A), 1x96wellsWash Buffer Concentrate, 20X, (Item B), 1x25mlStandards (Item C), 2x vialsAssay Diluent A (Item D), 1x30mlAssay Diluent B (Item E), 5X, 1x15mlDetection Antibody MMP-8 (Item F), 2x vialsHRP-Streptavidin Concentrate (Item G), 160X, 1x200ulTMB One-Step Substrate Reagent (Item H), 1x12mlStop Solution (Item I), 1x8mlStorage and Stability:Store powder at 4°C liquid at -20°C.
