Atrial natriuretic peptide (ANP), atrial natriuretic factor (ANF), is a powerful vasodilator, and a polypeptide hormone secreted by heart muscle cells. ANP is a 28-amino acid peptide with a 17-amino acid ring in the middle of the molecule. The ring is formed by a disulfide bond between two cysteine residues at positions 7 and 23. ANP is closely related to BNP (brain natriuretic peptide) and CNP (C-type natriuretic peptide), which all share a similar amino acid ring structure. ANP is secreted in response to stretching of the atrial wall, reduced Sympathetic stimulation of betaadrenoceptors, hypernatremia, Angiotensin-II, Endothelin and exercise. It is released by muscle cells in the atria of the heart in response to high blood volume. ANP acts to reduce the water, sodium and adipose loads on the circulatory system, thereby reducing blood pressure. ANP has exactly the opposite function of the aldosterone secreted by the zona glomerulosa in regard to its effect on sodium in the kidney.Sample Type:Serum and plasmaIntended Use:The kit is a competitive enzyme immunoassay for the in vitro quantitative measurement of ANP in rat serum, plasma; and other biological fluids not being tested.Sensitivity:15pg/mlRange:4.9-10,000pg/mlSpecificity:This assay has high sensitivity and excellent specificity for detection of rat ANP.Precision:Intra CV<10%; Inter CV <15%Test Principle:The immunoplate in this kit is pre-coated with secondary antibody and the nonspecific binding sites are blocked. The secondary antibody can bind to the Fc fragment of the ANP antibody whose Fab fragment will be competitively bound by both biotinylated ANP peptide and peptide standard or targeted peptide in samples. The biotinylated ANP peptide interacts with streptavidin-horseradish peroxidase (SA-HRP) which catalyzes TMB substrate solution. The intensity of the yellow is directly proportional to the amount of biotinylated ANP peptide SA-HRP complex but inversely proportional to the amount of the peptide in standard solutions or samples. A standard curve of known ANP concentration can be established accordingly. The unknown ANP concentration in samples can be determined by extrapolation to this standard curve.Kit Components:Assay plate (12x8 coated Microwells)-1Primary antibody-2x6uLStandard peptide-2x10uLBiotinylated peptide-2x10uLHRP-Streptavidin concentrate-1x60uLAssay diluent (5x concentrate)-1x15mLWash buffer (20x concentrate)-1x25mLTMB substrate-1x12mLStop solution-1x8mLAdhensive strip-2Instruction manual-1.Storage and Stability:Store powder at 4°C liquid at -20°C.
