Leptin is an identified protein product of the mouse obese gene. cDNA clones encoding Leptin have been isolated from human, simian, mouse, and rat cells. Rat Leptin shares approximately 96% and 82% sequence identity with the mouse and human protein, respectively. Rat Leptin cDNA encodes a 167 amino acid residue protein with a 21 amino acid residue signal sequence that is cleaved to yield the 146 amino acid residue mature proteinSample Type:Serum and plasmaIntended Use:The kit is a sandwich enzyme immunoassay for the in vitro quantitative measurement of leptin in rat serum, plasma; and other biological fluids not being tested.Sensitivity:69pg/mlRange:163.8-40,000pg/mlSpecificity:This assay has high sensitivity and excellent specificity for detection of rat leptin.Precision:Intra CV<10%; Inter CV <15%Test Principle:This assay employs the fast quantitative sandwich enzyme immunoassay technique. Antibody specific for rat leptin has been pre-coated onto a microplate. Standards, samples and rat leptin mix are pipetted into the wells and any Leptin present is bound by the immobilized antibody. Following a wash to remove any unbound reagent, a TMB substrate solution is added to the wells and color develops in proportion to the amount of leptin bound in the initial step. The color development is stopped and the intensity of the color is measured.Kit Components:Assay plate (12x8 coated Microwells)-1Standard-2x10uLMouse leptin Mix-4x8uLAssay diluent (5x concentrate)-1x15mLWash buffer (20x concentrate)-1x25mLTMB substrate-1x12mLStop solution-1x8mLAdhensive strip-2Instruction manual-1.Storage and Stability:Store powder at 4°C liquid at -20°C.
