Tumor necrosis factor alpha (TNF-a) is a pleiotropic molecule that plays a central role in inflammation, immune system development, apoptosis, and lipid metabolism. TNF-a is also involved in a number of pathological conditions including asthma, Crohn’s disease, rheumatoid arthritis, neuropathic pain, obesity, type 2 diabetes, septic shock, autoimmunity, and cancer. Rat TNF-a is synthesized as a 26kD type II transmembrane protein that consists of a 35 amino acid (aa) cytoplasmic domain, a 21aa transmembrane segment, and a 156aa extracellular domain (ECD).Sample Type:Serum and plasmaIntended Use:The kit is a sandwich enzyme immunoassay for the in vitro quantitative measurement of TNF-alpha in rat serum, plasma; and other biological fluids not being tested.Sensitivity:49pg/mlRange:81.9-20,000pg/mlSpecificity:This assay has high sensitivity and excellent specificity for detection of rat TNF-alpha.Precision:Intra CV<10%; Inter CV <15%Test Principle:This assay employs the fast quantitative sandwich enzyme immunoassay technique. Antibody specific for Rat TNF-alpha has been pre-coated onto a microplate. Standards, samples and rat TNF-alpha mix are pipetted into the wells and any TNF-alpha present is bound by the immobilized antibody. Following a wash to remove any unbound reagent, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TNF-alpha bound in the initial step. The color development is stopped and the intensity of the color is measured.Kit Components:Assay plate (12x8 coated Microwells)-1Standard-2x10uLRat TNF-alpha Mix-4x9uLAssay diluent (5x concentrate)-1x15mLWash buffer (20x concentrate)-1x25mLTMB substrate-1x12mLStop solution-1x8mLAdhensive strip-2Instruction manual-1.Storage and Stability:Store powder at 4°C liquid at -20°C.
